Research Abstract |
We examined the biosynthesis of blood group substances in tissues and obtained the following results: 1) The chemical structure of carbohydrate chains of blood group substances which exist in formalin-fixed and paraffin-embedded human and animal tissues, was determined by both lectin staining and immunological staining methods using exo- and endo-glycosidase. Distribution of isoantigenic substances, A, B and H in the tissue-sections was also determined by the methods mentioned above. 2) Biosynthesis of blood group substances was very active in cancerrous tissues of large intestine, but cancerous epithelial and acinal cells did not produce any blood group substances. These results indicate that A and B gene are inactivated by endogenous and exogenous factors. 3) We made experimental animal model for proof of mosaic and chimera phenomena of blood group substances. The existance and distribution percentage of different antigens (blood group substances) in tissues were examined by use of the model. We can also examine in what step abnormal chromosomes were produced during tissue differentiation model. 4) Our methods which mentiond in (1) are useful for diagnosis of accidental cases of blood transfusion.
