Research Abstract

1. The antitumor effect of ultrasound (US) and the enhancement of it by hematoporphyrin (Hp), protoporphyrin, pheophorbide, adriamycin (ADM) and FAD104 were observed using cell suspensions of AH130 and sarcoma 180.2. Histidine, a singlet oxygen scavenger, inhibited the antitumor effect of US with or without Hp or ADM, while superoxide dismutase and mannitol did not. 3. In ESR study, the generation of singlet oxygen by US was shown using 4-oxo-TEMPO. Hp or ADM with US increased ESR signals. 4. Sonoluminescence spectrum was observed when a saline solution was irradiated. It was not affected by active oxygen scavengers, indicating it is caused by the high temperature generated by cavitation. 5. In vivo animal experiments, US irradiation to AH130 of rats inhibited the growth of tumor and prolonged the life of host rats. Hp administered to host rats increased the antitumor effect of US. ADM and FAD104 also increased the US effect on sarcoma 180 and colon 26 of mice. 6. A phased array US transducer that can focus US spatially was tested to irradiate the rat liver in vivo. A histological damage was observed only at the focus of US, but not at the surrounding region.
