Research Abstract

細胞の増殖に関しては、EGFの受容体を過剰に発現しているA431細胞で、この受容体の刺激の結果としての増殖の亢進、抑制とともにG蛋白質を介して行われることを実証した。成熟ラットの肝細胞はα_1アドレナリン作動性受容体のみで交感神経の支配を受けるが、初代培養系に移し、低細胞密度で細胞を増殖させると急速にβ_2受容体が発現してくる。この時β_2受容体刺激はG_sを介してcAMPを産生させ、この情報がG_1期の後期に与えられれば、EGF、インシュリンの存在で細胞はS期に入る。一方、増殖を抑えるために高細胞密度で初代培養を行うとα_1受容体を介する応答が継続する。この時もEGF、インシュリン共存下で、この受容体刺激は多くの細胞をS期に移行させるが、この刺激(G_δを介するホスホリパ-ゼC活性化とそれに続くCa^<2+>動員)はG_0→G_1の初期1〜2時間でよい。細胞の分化に関してはHL60細胞を2時間、単独ではこの細胞を分化させない濃度のビタミンA、活性型ビタミンD_3、グルココルチコイドで核内受容体を刺激すると、同様に単独では細胞を分化させないINFδ、TNFα、PGE_2、PGF_2αに応答して細胞は分化した。これら細胞膜受容体刺激の効果は百日咳毒素によって完全に消失し、G蛋白質の関与が証明された。すなわち、核内受容体を介してある種の遺伝子が発現して、G蛋白質を介する細胞内情報経路が開くと考えられる。事実、これら細胞膜受容体刺激は核内受容体刺激後の細胞に限ってアラキドン酸遊離を惹起した。生物の進化とG蛋白質の関連については、線虫の細胞中にGi、Goの共通配列ペプチドに対して作製した抗体によって認識されるG蛋白質が存在し、一方アデニレ-トシクラ-ゼ活性がGTP結合蛋白質を介して調節されることを見出した。細胞性粘菌ではcAMP受容体がGTP結合蛋白質を介してアデニレ-トシクラ-ゼを活性化する。これら下等生物におけるGTP結合蛋白質の同定が今後の課題である。
