Research Abstract

The rat pancreatic beta cell transformed by simian virus 40 (SV-PB 1205) were transplanted into the spleen or subcapsular region of kidney of diabetic rats. Since the SV-PB1205 cells implanted could not find in situ in spite of preserving plasma glucose level of diabetic rats to normal for at least two weeks, insulin was thought to be originated from destructed SV-PB 1205 cells.
SV-PB 1205 cells seeded on the dextran beads (cytodex 3) could survive or propagate, however, those encapsuled in the arginate-polylysine-arginate microcapsules could not. A new pancreatic beta cell line, which can grow under various environments, is expected.
Human pancreatic beta cell, insulinoma cell and glucagonoma cell were cultured in vitro or transformed by plasmid (PMT10D), however, any continuous cell line from those cells have not been established as yet.
For an effort to create of hybrid type artifical organ, transformed rat hepatic cell line (RTH33) was established by transfection of PMT10D. It was further evidenced that the RTH 33 cell line preserved hepatocyte specific functions such as synthesis of urea and glucose and conjugation of bilirubin in vitro.
