Retinoblastoma (RB) is one of the most common intraocular malignant tumor. Recently with the analysis of the pedigree, chromosomes, and DNA, it is thought that the lack of a depressor gene, RB gene, produce RB tumor. With cell fusion technique, we confirmed that RB is induced by the lack of gene(s). RB Y79 cell line was fused with normal human fibroblasts, and the hybrids between them were examined microscopically. Those were all fibroblastic in appearance and grew well on the culture dishes and not well in suspension. So it was confirmed that RB is induced by the lack of gene(s). We also tried to do cell culture of RB from the three clinical cases, and in one case, we succeeded in culture. Two types of the cells were seen, the one was fibroblastic and showed two layer growth partly. The other was with many processes, and similar to a nerve cell. The latter had a tendency to stop growing and decreased in number.
