Research Abstract |
1) An IgAl-binding lectin, Jacalin, was isolated from the seed of Artocarpus heterophyllus. Jacalin is a glycoprotein with two subunits. Interaction between Jacalin and human immunoglobulins were studied by precipitation and latex agglutination assay. Jacalin bind selectively IGAI, but not other class or subclasses of immunoglobulins. Jaclin bind Gal_<beta>l-3GalNAc and GalNAc in IgAl hinge region. 2) The separation of IgAl and IgA2 subclasses were easily achieved by Jacalin affinity affinity chromatography. This method has several advantages compared to antibody affinity chromatography. 3) Normal serum IgA and secretory IgA of subclass IgAl were isolated from pooled serum and milk. They were tested for their susceptibility to bacterial IgA preteases from H.influenzae, Str. pneumoniaer N.Gonorrhoeae, and N. Meningitidis. Both serum and secretory IgAl exhibited resistance to IgA proteases. The resistance of sIgAl and IgAl were found to be due to their enzyme neutralizing antibody activity

, because the Fab fragment but not the Fc of sIgAl showed inhibitory activity. The IgA protease-ndutralizing antibody activity present in secretory IgAl on mucosal surfaces as well as in serum IgAl in the blood play an effective role against pathogenic bacterial infections in human. 4) We collected so far 14 samples of Jackfruit's seeds, including 5 species of Artocarpus, from 9 countries. Lectins from Artocarpus integer(Sao Paulo,Singapore), A.lakoocha(West Bengal), A. tokinesis(North Vietnam) and A.communis(Nigeria) selectively bind human IgAl and 4 out of 5 IgD samples, but not IgA2m(1), IgA2m(2), IgG, IgM, IgE, free secretory component. On the contray, Jacalins from Artocarpus hererophyllus(9 samples from manila, Okinawa,Sao Paulo Singapore, West Bengal, La Reunion and South Africa) showed different immunoglobulins binding patterns which are classified into 3 types. (1) Jaclins from Okinawa and La Reunion bind IgAl, IgA2m(2), IgM, IgD, IgE. FSC,SIGA, but not IgA2m(l) and monomer IgG. (2) Two sample of Jacalins from manila bind FSC, sIgA, and all classes of Immunoglobulis except gor IgG. (3) Jacalin from south Africa, West Bengal, Sao Paulo, Singapore, one batch from manila bind IgAl and 4 out of 5 IgD samples, but not other classes of immunoglobulins. Less
