Outline of Annual Research Achievements

タンパク質は、骨や動物遺存体などの生物由来の発掘試料だけでなく、壁画や絵画などの彩色材料、毛皮で作られた衣類、更には食物残滓が付着した土器など多岐に渡る文化財に残存する素材であり、古代DNAと同様に重要な研究対象である。本研究では、発掘又は保存環境により汚染された試料を含む文化財から採取した極微量片に残存する有機物（特に、古代タンパク質・ペプチド）を効率的に抽出し、高分解能質量分析を用いたアミノ酸配列情報解析を行うことで、西アジアを中心に収集した各文化財に含まれる当該有機物の特定を可能にする手法を確立する。
これまでに、ナノ液体クロマトグラフィーによる分離とマトリックス支援レーザー脱離／イオン化（MALDI）質量分析もしくはエレクトロスプレーイオン化（ESI）質量分析を併用することで、不純物を含みかつ極微量な文化財試料からも古代のタンパク質が抽出・分析可能であることを示すことに成功しており、その成果として、J. Paul Getty美術館が所蔵するローマ期エジプトの三連祭壇画（A.D. 180～200）やエジプトのナガ・エッデイル（Naga ed-Deir）の墓から発掘された箱型木棺（およそ2,300 B.C.）の彩色に用いられた膠着材の原料として膠（コラーゲン）が使われていること明らかにした。また、アフガニスタンのバーミヤーン遺跡の東・西大仏（A.D. 400～600）のそれぞれから採取された極微量の彩色片から、コラーゲンや乳タンパク質として知られるカゼインを検出することに成功した。特に、バーミヤーン遺跡の彩色に用いられた膠は、ウシのI型コラーゲンのアミノ酸配列と一致しており、僅かなアミノ酸配列の違いからヤクなど近縁の動物種と区別することが可能であった。これらの成果は、本手法により、広範な年代・地域に渡る文化財中のタンパク質分析が可能であることを実証する結果である。
