生体分子応答性ゲルを用いたマイクロアクチュエータシステムの構築

Outline of Annual Research Achievements

本研究では,生体分子応答性ゲルのコンセプトを拡張し,標的分子を入力としてインプットすると動的架橋のネットワーク構造変化により情報変換し,マクロな体積変化を出力情報としてアウトプットできるスマートゲルシステムの創出を試みる。平成28年度は以下のような研究成果が得られた。
(i)屈曲する生体分子応答性ゲルの合成とマイクロアクチュエータシステムの構築:平成27年度と同様にポリアクリルアミド(PAAm)ゲル層の片面上で,リガンドとしてシクロデキストリン(CD),鋳型分子としてビスフェノールA(BPA)を用いた分子インプリント法によりBPA応答性ゲル層を形成させた。この2層ゲルの構造を最適に設計したところ,BPA水溶液中で一方向に曲がる明確な屈曲挙動を示した。したがって,BPAに応答して屈曲する分子応答性ゲルの合成に成功した。
(ii)膨潤収縮する生体分子応答性ゲルの合成とマイクロアクチュエータシステムの構築:光学顕微鏡を用いてマイクロ流路内に応答膨潤型のグルコース応答性ゲルと応答収縮型のBPA応答性ゲルを調製した。前者のグルコース応答性ゲルは,グルコース存在下で速やかに膨潤し,その応答挙動は動的架橋として導入したレクチン-糖複合体の含有率によって変化した。一方,後者のBPA応答性ゲルは,BPAに応答して収縮した。マイクロ流路にグルコースあるいはBPAを流すと,グルコース応答性ゲルでは流量が減少し,BPA応答性ゲルでは流量が増加した。したがって,グルコース応答性ゲルは標的分子に応答して自律的にクローズするマイクロバルブ,BPA応答性ゲルはオープンするマイクロバルブとして利用できることがわかった。したがって,生体分子応答性ゲルは自律応答型マイクロ流路制御バルブとして利用できることが明らかとなった。
