Outline of Annual Research Achievements

「地上に降り注ぐ太陽光から有益なエネルギー源となる物質を生産する」という人工光合成の実現は、光化学系II (Photosystem II,PSII)の立体構造(Umena, Kawakami, Shen, Kamiya. Nature (2011))解明により、ますます現実味を帯びてきた。分子構造がわかれば原子間相互作用を解析することでその分子が行う化学反応の様子も見ることができるからである。一方で得られた結晶構造を眺めるだけの研究手法では理解に限界がある。本研究では、PSII結晶構造を分子化学の立場で、理論化学的手法を用いて解析することで、PSII水分解・酸素発生反応機構の解明を目指した。
PSIIのプロトン移動経路と基質水分子取り込み経路は、蛋白質内部のチャネル空間に存在する水分子ネットワークで構成されている可能性が高い。そこで、今年度はPSII蛋白質の触媒部位Mn4CaO5と蛋白質表面を結ぶチャネル空間を全て明らかにした。このチャネル空間は、シアノバクテリア由来のPSII結晶構造と高等植物由来のPSII結晶構造の両者で解析した。
また、PSIIのプロトン移動は電子移動と連動して起こる。電子移動経路のエナジェティクスも明らかにするため、モデル蛋白質の電子移動経路中のコファクター酸化還元電位を計算することに成功した。今後、PSIIでの電子移動経路におけるコファクターの酸化還元電位を解析する上で重要な知見を得ることができた。
