Outline of Annual Research Achievements

大気圧プラズマは照射すると特に液中に大量の活性酸素種(ROS)を生成することが知られ、がん治療への応用も研究されている。本研究では、これらの大量のROSを細胞内に導入する手法として、温熱に注目して併用効果を調べた。温熱処理、即ち温度上昇は細胞膜の透過性を亢進し、併用により細胞内へのROS移動が亢進すると思われる。大気圧プラズマ照射実験に際しては、NUグローバル社製のHeプラズマ発生装置を用いた。周波数60Hz, peak-to-peak電圧は7 Kvを用いた。Heガス流量は2 L/min であったアポトーシスについては、細胞膜表面上へのフォスファチジルセリンの発現DNA断片化、形態変化、関連タンパク質の発現等を指標に検討した。その結果、単独での影響がほとんど認められない42度,20分をプラズマ処理後に使用した場合、顕著な増強効果が認められた。
大気圧プラズマは大量の活性酸素種(ROS)を生成することは判明しているが、その詳細な機序について、特に発生場所については不明な点が多い。そこで、揮発性アルコール類を用いて、プラズマに特異的なフリーラジカルが生成するか否か、スピン捕捉剤としてDBNBS(3,5-dibromo-4-nitoroso benzene sulfonate)を用いESR-spin trap法にて検討した。作製した大気圧非平衡アルゴン(Ar)プラズマ照射装置を使用、条件は印加電圧Vp-p:18 kV、周波数:20 kHz、ガス流量:2 L/minである。その結果、用いたアルコール類ではOHラジカルにより生じる水素引き抜き反応によるラジカル種のみが認められた。大気圧プラズマの化学反応の特徴は多量のOHラジカルに起因すると思われる。
