Outline of Annual Research Achievements

本研究は新学術領域公募班におけるH25-26年度の研究成果の一つである電界誘起気泡を発展させマイクロプラズマ・液体界面による遺伝子導入や癌細胞アポトーシス等を目的としたバイオメディカル分野に貢献する実用化デバイスの製作,及びプラズマ活性種凝集を利用した新機能材料生成への応用研究を目標として研究を行いました.
具体的な研究成果として,申請者の発明であるプラズマ誘起気泡を用いたダイナミックレンジの大きな固さを有する対象への穿孔・試薬導入技術を確立し,さらに独自開発であるプラズマ活性種を凝集・循環する機能を持った3次元循環流路を用いることにより循環中の流路内に存在するプラズマ活性種を含む溶液を利用してたんぱく質結晶生成に成功しました.このプラズマ誘起気泡によるタンパク質結晶生成において,結晶品質のX線解析を行いコントロールである蒸気拡散法の条件と変わらない品質であることも確認しました.
さらにこのプラズマ気泡を利用した3次元循環流路を用いることにより,気液界面の活性種を利用して低侵襲かつハイスループットな細胞への遺伝子導入へ応用できることもわかりました.このデバイスを利用することにより,これまで遺伝子導入が困難であった酵母や藻類細胞への遺伝子導入への可能性が高まり,基礎実験結果ではその実現性を大きくサポートできる結果が出たことにより今後の展開が期待される新しい遺伝子導入技術が生み出される結果となりました.
このような高負付加価値膜面の生成技術の開発により,バイオ・電子・化学・材料等の広い分野に貢献する可能性が広がりました.
