Outline of Annual Research Achievements

本研究の目的は、真核生物の鞭毛・繊毛の微小管に結合するタンパク質がなぜ決まった位置にしか結合しないのか、その特異的位置決定の分子機構を解明することにある。
真核生物の鞭毛・繊毛は生物種を越えてよく保存された9+2構造をもつ。9組の周辺微小管が2本の中心対微小管を取り囲んだ円筒状の構造をしている。各周辺微小管の上の決まった位置にモータータンパク質ダイニンの外腕と内腕が長さ方向に周期的に結合している。このダイニンが隣接する微小管に対してATP依存的に滑り運動を行うために鞭毛は波打ち運動を行う。この運動は、微生物や精子の運動だけでなく、人体における脳室、気管上皮、輸卵管上皮などにも存在し、ヒトの健康に重要な役割を果たしている。ダイニンが決まった位置に周期的に結合していることが運動形成において重要であると考えられるが、この位置決定メカニズムは不明である。
我々は、鞭毛研究のモデル生物である緑藻クラミドモナスを用いて、外腕ダイニンの位置決定に関わるダイニン外腕ドッキング複合体と相互作用するタンパク質を探索することでその解明を目指した。微小管上でドッキング複合体サブユニットと結合する2つのタンパク質を化学架橋法と免疫沈降法によって同定した。これらのタンパク質の局在を調べたところ、微小管内部であった。近年、クライオ電顕技術の発展に伴って微小管内部微細構造が明らかになってきたが、2つのタンパク質はその実体であった。我々はこれらのタンパク質が微小管の内側から微小管上の構造の位置を決定している仮説を立てた。しかし、このタンパク質の欠損株はダイニンは野生株と同様に存在しており、残念ながら位置決定には関わらないことが明らかになった。一方で、興味深いことに、このタンパク質の欠損株の微小管は物理的刺激によって分解しやすいことが判明した。この新規タンパク質は微小管安定化に関わるという副産物的な知見を得た。
