Outline of Annual Research Achievements

NOP1の標的タンパク質や制御タンパク質を単離することを目的として、NOP1結合タンパク質を探索し、得られた候補遺伝子について機能欠損変異株を作出し、表現型を解析することで関連遺伝子の同定を試みた。まずNOP1結合タンパク質を同定するため、U-boxドメインを欠失した不活性型NOP1-Citrineを発現する形質転換体を用いて免疫沈降を行い、沈殿物に対して質量分析を行った。その結果、数百に及ぶNOP1結合タンパク質候補が単離された。得られたNOP1結合タンパク質候補に対して様々な情報をもとに、細胞間隙形成と関連のありそうな候補を絞り込んだ。選抜したNOP1相互作用因子候補には微小管や細胞骨格関連因子、セルロース合成酵素関連因子、膜交通関連因子、受容体キナーゼなどが含まれていた。これらのNOP1相互作用因子候補についてCRISPR/Cas9ゲノム編集システムによって機能欠損株の作出を行い、表現型の観察を行ったところ、複数の機能欠損株で基質形成に異常が確認され、NOP1と協働して気室形成に関与する可能性のある因子が複数単離された。その中でも、微小管とセルロース合成酵素の双方に結合して働くことが示唆されているCSI1(CELLULOSE SYNTHASE-INTERACTIVE1)遺伝子のオーソログに着目し、更なる解析を行った。その結果、ゼニゴケCSIの機能欠損株では、気室形成密度が低下すること、気室形成が起こる領域でも形態の異常な気室や気室孔が形成されることが明らかとなった。このことからCSIは正常な気室形成に必要な因子であると考えられ、CSIは微小管と細胞壁合成酵素の制御を介して気室形成を正に制御することが示唆された。
