Outline of Annual Research Achievements

成長が盛んな細胞では、葉緑体遺伝子と細胞壁遺伝子はともに活性化され、免疫応答などのストレス応答に伴って同時に抑制される。この事実は、両者を同調するメカニズムの存在を示唆するが、その実体は不明である。一方，色素体はレトログレードシグナル(RS)を発信し、葉緑体機能と細胞全体の遺伝子発現プログラムを同調させている。本研究は，これまで知られていない葉緑体と細胞壁の同調メカニズムの解明を目指し、ミトコンドリアを含めたオルガネラ間ネットワークによる細胞壁制御に注目し、研究を実施した。
その結果、光合成によるエクスパンシン遺伝子の発現制御について、光合成阻害剤のDCMUやDBMIBによって、エクスパンシン遺伝子が発現抑制を受けることを明らかにした。これらの研究から、ストレス下での成長抑制に、葉緑体レトログレードシグナル(RS)が関わっている可能性が示された。
また、DBMIB処理によって、キシログルカン転移酵素酵素などの細胞壁調節酵素の発現が誘導される現象を見出し、ストレスによるミトコンドリア機能傷害によってミトコンドリアRSが生じ、細胞壁調節酵素などの遺伝子発現が制御されていること、ミトコンドリアRSにCa2+シグナルが関係する可能性を示した。さらに、ストレス時にアポプラストで生じるROSがシグナルとして葉緑体制御に関わる可能性についても検討を進め、細胞外に与えたH2O2が、長く続く葉緑体内Ca2+濃度上昇を引き起こすことなどを明らかにした。
