Outline of Annual Research Achievements

ヒトにおけるストレス感受性には個人差があることが分かっており、その背景には環境要因に加えて遺伝的要因も強く関わっていると考えられている。日本産野生マウス系統であるMSMは、実験用系統であるC57BL/6J (B6)と比較して、極度に高い不安様行動と恐怖反応を示すことから、その遺伝的基盤を解明することで、ヒトのストレス感受性や不安の個人差に関わる遺伝的基盤の理解に結びつけられるのではないかと期待される。詳細な遺伝学的解析により、その責任遺伝子が、動物のストレス応答に重要な役割を果たす下垂体アデニル酸シクラーゼ活性化ポリペプチド（PACAP）をコードするAdcyap1遺伝子であることを見出した。MSMとB6系統由来のPACAP遺伝子は、タンパク質コード領域には多型が無いが、視床下部領域における発現量はMSMに由来する遺伝子で有意に高く、またPACAP遺伝子転写産物のスプライシングパターンにもMSMに特徴的なスプライシングバリアントの増加がみられた。バリアントの5’非翻訳領域にレポーター遺伝子であるルシフェラーゼ遺伝子をつないで行った発現解析の結果、MSMタイプのバリアントが最も高い翻訳効率を示すことが分かった。更にこのPACAPの高発現とスプライシングパターンには遺伝子イントロン内の２塩基繰り返し配列多型が関与していることが分かった。そこで、ゲノム編集によりB6にMSM型の変異を導入すると、遺伝子発現パターンがMSM様に変化することが分かった。このことから、PACAP遺伝子内の２塩基繰り返し配列多型により高発現とスプライシングの変化が生じ、それによりPACAPの増加が生じることで不安様行動が増加することが分かった。また、このPACAP髙発現マウスでは、恐怖条件付け学習後の恐怖記憶の消去が効率よくできないことから、PACAPの高発現とPTSDとの関連を指示する結果が得られた。
