Outline of Annual Research Achievements

血管内皮細胞に分化するときと分化した内皮細胞が活性化するときのエピゲノム変動を包括的に探索し、転写サイクルを解析した。まず未分化 ES 細胞が VEGF 刺激を受けて内皮細胞に分化する際の whole transcriptome とヒストン修飾変化の ChIP-seq データをまとめ、内皮分化に必要な転写ネットワークと、必須転写因子において、転写のブレーキヒストンマークからアクセルヒストンマークにスイッチしていく原理を解明した。さらに必須転写因子をノックダウンすることで、内皮分化が妨げられるだけでなく、内皮分化系では認められない他系列分化に関与する因子が誘導され、分化システムが破綻することが判明した (Nucleic Acids Research, 2017 In press)。
一方、分化した内皮細胞が VEGF 刺激を受けて血管新生促進の方向に活性化するシステムも whole transcriptome やヒストン修飾変化を時系列でもって動的変化を追跡したところ、血管新生に必須な急性期転写因子群の制御領域には H3K4me3 転写アクセルマークと H3K27me3 転写ブレーキマークが共存する bivalent 状態であること、エピゲノムレベルでのブレーキは PRC1, 2複合体が担っているが、 VEGF 刺激を受けて一過性に活性化する際には、内皮に特有な PRC1 バリアントが血管新生転写因子の制御領域に15分後に集積し、 PRC2 の抑制能解除と転写アクセルマーク H3K4me3 の修飾を向上させるトリソラックス複合体のアダプター因子が集積してくること、この段階で PolII の転写が行われるが、PRC1 のconventional タイプにその後引き継がれることで、再度 H2AK119 のユビキチン化が誘導され、転写がストップする新たな現象を見出した。
