Outline of Annual Research Achievements

酸素応答は酸素を利用する生命システムにおいて最も重要な環境適応機構の一つである。脊椎動物における低酸素応答の分子機構はそのマスター転写因子であるHypoxia inducible factor (HIF-) を中心に研究が展開されてきた。酸素応答がすべてHIF-の制御のみで説明できないことや、酸素分子が関わる生命現象の多様性から、酸素応答に関わる生命システムには未知の領域が多く残されていることが推定され、その全体像の解明が重要な課題である。本研究では、独自に構築した様々なプロテオーム解析技術やロボット実験技術を駆使して、主にタンパク質水酸化に着目した解析を行うことで、低酸素応答の全体像解明を目指す。
まず、水酸化ターゲットタンパク質の候補を情報生物学的手法により選択し、エピトープタグを付加した候補タンパク質を発現させ、高精度質量分析法にて詳細な翻訳後修飾の解析を実施する、Target Hydroxylome Scan法を構築した。本方法をヒト型ロボットを用いて大規模に実施する系を構築した。しかしながら、質量分析計による水酸化ペプチドの同定には試料調製過程で生じる不均一なメチオニン酸化反応が大きな障害となることが判明したため、すべてのメチオニンを強制的に酸化させる手法を開発し、プロリンなどの水酸化の同定確度が改善した。また、水酸化ペプチドを定量するための、ターゲットメソッドも構築した。また、各種水酸化酵素と相互作用するタンパク質を同定するための手法として、Biotin-tagの導入による相互作用解析法やBioID法による一過的相互作用検出法を確立した。さらに、独自に開発した次世代型定量プロテオミクスiMPAQT法を駆使して低酸素応答時の代謝酵素の変化を網羅的に精密定量することに成功した。
