Outline of Annual Research Achievements |
〔目的〕微小管結合たんぱく質タウは、加齢依存性神経変性疾患脳で異常な修飾を受けて蓄積し、神経細胞死を引き起こす。しかし、疾患脳でタウの異常代謝がどのように進行するのかは不明である。疾患脳ではタウは多くのリン酸化を受けるが、中でもSer262のリン酸化はタウの異常代謝を引き起こし毒性を増加させる。本課題では、Ser262∙356でリン酸化されたタウの生成・代謝に関与する遺伝子を網羅的に同定することを目的とした。〔方法〕優れた遺伝学的モデルシステムであるショウジョウバエタウ毒性モデルを用い、Ser262でリン酸化されたタウの量を制御する遺伝子をRNAiによるノックダウンを行いてスクリーンした。タウの量とリン酸化レベルはWestern blottingで定量し、タウの毒性について切片を作成して神経細胞死によって起きる空包の大きさを測定した。〔結果および考察〕これまでにリン酸化タウの量と毒性に関わる遺伝子二つを新規に同定し、そのメカニズムを調べている。一つはE3 ubiquitin ligase, UBR4ドメインを持つタンパク質であり、リン酸化タウまたはそのリン酸化酵素のタンパク分解に関与する可能性がある。もう一つの遺伝子はS6Kのショウジョウバエホモログであり、そのノックダウンによってリン酸かタウの量が増加する。S6Kはインスリンシグナリングの下流に位置する。インスリンシグナリングの乱れと認知症の関連が指摘されており、この研究を発展させることで、インスリンの下流にあるシグナルがタウの毒性を変化させる分子メカニズムが明らかになる可能性がある。〔結論〕本研究から、タウ毒性に関わる新たな経路が複数同定された。将来これらの知見をタウオパチー治療戦略の開発に繋げたい。
