Outline of Annual Research Achievements

SOD1遺伝子変異を原因とする筋萎縮性側索硬化症(SOD1-ALS)では、ミスフォールド型の変異SOD1タンパク質が病変部位である脊髄運動ニューロンに異常に蓄積する。しかし、ミスフォールド型SOD1の分子構造的特徴や毒性については明らかとなっていない。私たちはこれまでに、変異SOD1がジスルフィド結合でクロスリンクされた「S-Sオリゴマー」が疾患の発症に関わるミスフォールド型SOD1であることを報告してきた(PNAS 2006; JBC 2013)。本研究では、S-Sオリゴマーを特異的に認識する抗体の開発に成功し、病変部位である脊髄前角の運動ニューロンにS-Sオリゴマーが加齢に伴い蓄積することを見いだした(Mol Neurodegener 2017)。一方で、SOD1-ALSの非病変部位である小脳には、S-Sオリゴマーの形成・蓄積は認められなかった。また、SOD1遺伝子に変異を有するALS患者の脊髄には、本課題にて開発したS-Sオリゴマー特異的認識抗体によって検出されるSOD1が蓄積していることを見いだした。よって、病因性変異に伴って、SOD1の構造が不安定化することで、S-S結合の組換え現象が生じ、S-Sオリゴマーが形成することを示すことができた。しかし、S-Sオリゴマーが神経変性に関わる毒性を発揮するのかについては未だ不明のままである。そこで本研究では、S-Sオリゴマーを線虫に投与することでその毒性の評価を行った。その結果、S-Sオリゴマーを添加すると寿命短縮・運動性低下が見られたのに対して、正常型SOD1を投与しても毒性は確認されなかった。以上より、変異SOD1タンパク質はS-Sオリゴマーを形成することで病原性を発揮することを提案する。
