Outline of Annual Research Achievements

本研究では、in vivoでの動態機能解析、X線結晶構造解析およびクライオ電子線トモグラフィー法による構造解析、さらにin vitroでの生化学的機能解析法を上手く組み合わせることで、フック完成シグナルに伴う輸送装置の基質認識モード切り替え機構を原子レベルで解明することを目的としている。本年度の主な成果は以下に示す。
1. べん毛輸送シャペロンであるFlgNの立体構造を2.3Å分解能で決定した。構造に基づいて変異体解析を行った結果、α1ヘリックスとα2ヘリックスを繋ぐNループ領域がFlgNの輸送シャペロン活性を巧みに制御する構造スイッチとして機能することが判明した。
2. べん毛輸送シャペロンであるFliSとFlhAcとの間の強い相互作用を介してフラジェリンのC末領域がFlhAcに安定にアンカーされると、輸送ゲート複合体によってフラジェリンが効率よくアンフォールドされて細胞外方向へ輸送されることが判明した。
3. FliKのC末天然変性領域がフォールドすると、FliKのC末コアドメインが構造変化し、その結果輸送ゲート蛋白質FlhBに結合することが示唆された。
4. FlhAリング複合体形成は非常に高い正の協同性を示すこと、さらにその協同的なサブユニット間相互作用にはリンカー領域が重要であることが明らかとなった。
5. FlhAリング複合体が構造変化すると、輸送装置の基質特異性が切り替わることが判明した。
