Outline of Annual Research Achievements

本研究では、真核生物鞭毛の振動運動に着目し、原生生物から哺乳類に至るまで保存されている「9+2構造」の軸糸において複雑な運動活性を生み出すモータータンパク質ダイニンの高度に組織化された制御機構の解明を目指した。具体的には、独自の手法で、ダイニン分子の昨日の自律性を保ったまま複雑性を捉え、さらに、このダイニン分子の自律的複雑が統合され、秩序系としての鞭毛の振動運動に至る時空間展開機構の理解を目指した。前年度及びその繰越の計画の実績によると、自律性を保ったまま複雑性を捉えるには、ダイニンと相互作用している微小管の挙動の解析がまず有用である。この系では、振動誘導は非常に難しいが、外部から当てた力学的変化に対するダイニンの応答を着実に捉えることができた。抽出精製したダイニンにより誘導される微小管の滑り運動に外部からの力学的変形が安定したシグナルとして作用しうることは驚きであった。もう一つの手法は、滑り運動が秩序だった振動運動として時空間制御される過程を詳しく解析することを目指したもので、膜除去後の精子を2.5 microM以下のATP条件下に置き力学的変形を与えて振動誘導条件を探索した。最終年度の最大の成果は、自律的運動が起こる域値濃度以下のATP下では、ATPによらず、力学的効果により振動が安定して起こるらしいことを見出した点にある。観察を行うチャンバーの底面からの距離を変えて鞭毛運動の変化を誘導できるかに挑戦した結果、表面張力の影響を受ける境界条件において、この現象が見られるらしいことがわかった。このことは、この条件以外の時には、振動の制御は、ATPに依存した、つまり微小管のすべり速度に依存した制御を受けていると考えられるが、この境界条件では力学的効果(物理的特性)が優位性を持つことを意味する。
