Outline of Annual Research Achievements

光学・蛍光顕微鏡法を駆使する事で観察が可能な高分子微粒子をタンパク質をはじめとする生体物質のモデルととらえ、コロイドサイズの高分子微粒子が示す動的な秩序構造を発展させるために研究を進めてきた。
研究最終年度には、これまでに開発してきた、生体細胞様に高分子微粒子を構造化した中空体の動的秩序の検討を進めてきた。特に、様々な刺激に対して高速に応答するための微粒子の構造設計を行った。高分子微粒子の動的な秩序を構築する化学振動反応(化学基質濃度依存性、温度依存性)を詳細に検討した。従来用いていた高分子微粒子では、これらの条件を大きく変化させてしまうと微粒子が凝集してしまうという課題があった。そのため、耐塩性があり、そして高い温度でも凝集しないために新たに親水性モノマーを高分子微粒子に導入し、構造の最適化を実施した。その結果、化学振動反応の条件を、従来よりも広範にわたり変化させることが可能となり、その結果、数秒のオーダーで体積振動または分散凝集振動をする高分子微粒子を実現する事が出来た。これらの微粒子は、集積化後もその特徴を兼ね備えており、人間の心臓と同様の時間スケールで体積振動する事が可能な材料を得る事が出来た。すなわち、心筋細胞が集積化した心臓のように、ミクロな微粒子を集積化する事が、高機能発現に有意であることを実証する事が出来た。
以上の検討を大きく発展させるために、領域内共同研究を実施してきた。その中でも特に、複合化ハイドロゲル微粒子の膨潤状態の可視化をクライオ電子顕微鏡により達成し、用いる高分子微粒子の詳細な構造解析(特にトモグラフィー解析)を実現する事が出来た。また、水中における分子スケールでの高分子微粒子の可視化を原子間力顕微鏡により達成し、タンパク質等との相互作用に関する知見を得る事が出来た。
