Outline of Annual Research Achievements

本課題では,in-cell NMRを用いて,細胞内混雑環境下での生体分子の動的秩序の解明を目指した.特に周辺環境の影響を受けやすい天然変性蛋白質やマルチドメイン蛋白質を対象としている.
モデル試料として,本年度は,Drk蛋白質のNMR解析を行った.DrkのN末端ドメイン(Drk-N)は,希薄溶液下で,フォールド状態と変性状態が混在する性質を持つ.細胞内でのDrk-Nの動態を明らかにするため,AlaとLysの選択的アミノ酸標識を用いて信号を限定し,in-cell NMR測定を行った.解析の結果,Drk-Nは細胞内では,より変性状態に平衡が傾いているということが示唆された.また,Drkの3つのドメインの相対配置を明らかにするために,常磁性金属結合タグDOTA-M8-SPYを蛋白質に結合させ,擬似コンタクトシフト(PCS)の観測を行った.構造解析に有効なPCSの観測に成功した.現在,このデータの解析を進めており,希薄溶液下と細胞内環境下でのドメイン相対配置を比較する予定である.
別のモデル蛋白質であるRasは,13C標識したRasをヒト培養細胞に導入し,細胞内蛋白質のメチル基シグナルの観測に成功した.希薄溶液下と細胞内環境下でのNMRスペクトルの比較から,複数のピークで異なる化学シフト値が観測され,細胞内のRasの立体構造に何らかの変化が生じていることが示唆された.
また,生体分子の動的秩序の解明に有力な溶液NMR法と,その解析法に関する総説および申請者のいくつかの研究成果をまとめた記事を2報発表した (T.Ikeya et al, J. Comput. Chem. Jpn., 7, 65-75 and T.Ikeya et al, Biochim. Biophys. Acta, Gen. Subj., 1862 (2) 287-306).
