Outline of Annual Research Achievements

ベシクル系をベースとして細胞機能を再現するプロトセルを、非平衡ゆらぎが起こる場として考え、生命現象がどのような物理的機構を経て成り立っているのかを理解する。具体的には、ベシクルの内空間から新たに脂質を供給し自己成長・分裂を促すことで、ベシクルの形態変化を観察しその物理を解析する。平成29年度は以下のような研究実績を得ることができた。
再構築した脂肪酸合成系の最適化を行なった。具体的には酵素比を変えることにより、飽和脂肪酸メジャーな状態から不飽和脂肪酸をメジャーとする系に改良した。これにより、合成産物である脂肪酸による酵素タンパク質の凝集や不活化を防ぐことに成功した。改良した系により、C18:1、C16:0、C16:1をメジャー脂肪酸として合成することができた。また、嫌気条件下で強瀬を行なったところ、C18:1のみ約1.5倍の合成量の増加が観察できた。これらの最適化により、再構築系で200uMの脂肪酸を合成することができた。これは、直径20umのベシクル内部でどう反応を行なった場合、母ベシクルを構成する資質に対して約20%の脂質を合成できることを意味している。よりベシクル内部の環境に近ずけるため、また合成した脂肪酸の局在場所を提供する目的で、直径200nmのリポソームを反応液に加えて脂肪酸合成を行なった。その結果合成初速度が大幅に上昇する結果が得られた。これは、合成された脂肪酸がリポソーム膜に吸収(局在)され、酵素に付着する頻度が減少し、その結果脂肪酸による酵素阻害が低下したことで合成スピードが保たれたものと考えられる。
最適化した再構築系をベシクルに内包し内部で反応を行いながらベシクルを観察した。その結果、わずかではあるがベシクル表面から細いチューブ状の樹状のベシクルが観察された。この現象が内部の反応依存的なものかどうかを現在確かめている。
