Outline of Annual Research Achievements

APOBEC3(A3)タンパク質群に属するA3B、A3F及びA3Gによる脱アミノ化反応を、リアルタイムNMR法によって解析した。A3Gは長鎖のDNA中の標的配列に対する活性が高いが、A3Bは短鎖のDNA中の標的配列に対する活性が高い事が分かった。またA3GはDNA鎖中の5'端近くに位置する標的配列に対する活性が高いが、A3BはDNA鎖中の中央に位置する標的配列に対する活性が高い事も分かった。A3Gは、DNA上の標的配列以外の箇所に非特異的に結合後、DNA上をスライディングする事で、標的配列を効率的に見つけて脱アミノ化反応を生じる。しかしA3BはDNAへの低い結合能の為に、スライディングの最中に標的核酸を見つける前に、DNA鎖から解離してしまう。この違いによって、両タンパク質における鎖長依存性及び位置依存性の違いを合理的に説明できた。
細胞中における分子混雑下では、生体分子の構造と相互作用が、試験管内とは異なる可能性が指摘されている。この問いに答える為に、ヒト細胞中におけるタンパク質のNMRシグナルを観測し、研究が進められている。一方核酸に関しては、細胞への導入が困難であった。今回毒素タンパク質SLOを用いた物質の導入法を高度化する事でこの問題を克服し、ヒト細胞中における核酸のNMRシグナルを観測する事に初めて成功した。
DNAの光損傷に伴いサイクリンD1遺伝子の上流領域から転写された非コードRNAは、同遺伝子のプロモータに繋留され、TLS蛋白質をリクルートする。結合に伴いTLSは構造変化し、CBP/p300と相互作用してこれらのHAT活性を阻害し、転写を阻害すると想定されている。今回TLSと非コードRNAの局所的な相互作用をNMR法で明らかにすると共に、この相互作用よってTLS分子全体に引き起こされる大きな構造変化を、FRETとAFMによって捉える事に成功した。
