Outline of Annual Research Achievements |
本研究では、一般的な糖鎖構造のグリコシド結合のO原子をCHF基に置き換えた化合物の合成法開発、特に2,6-sialylgalactose構造とGal-β(1,3)-GalNAc構造のアナログを、効率的に合成する手法の開発に取り組んだ。 まず、2,6-グリコシド結合は、糖ドナーのアノマー位がアクセプターの6位と連結したものであり、1,6-グリコシド結合と同様である。そこで、まず1,6-連結型のC-グリコシドを、効率的に構築するため、分子間カップリング反応を基盤とし、さらにアノマー位をsp3炭素のまま連結する手法開発を目指した。昨年度までに、ラジカル反応を基盤とした原子移動型カップリング反応が進行しうることを見出していた。そこで、これを立体選択的に、またより高効率で進行させるドナーを探索した。その結果、α-選択的に2糖構造を構築できるドナーを見出すことに成功し、これを選択的なイソマルトースアナログの合成に適用することができた。また、立体選択性の改善は必要であるが、2,6-sialylgalactoseアナログも合成できた。一方で、同じ手法でCHF基の構築を検討したが、予想外にも反応効率の著しい低下が見られた。このことから、本手法でのCHF基構築は困難であり、別の手法開発が望ましいことがわかった。 一方で、1,3-グリコシド結合などは、原子移動型カップリングが適用しにくい結合様式であることから、sp3-sp2のクロスカップリング反応を適用することを考え、モデル反応を種々検討した。その結果、光レドックス触媒を利用して発生させたラジカル種と芳香族化合物のクロスカップリング反応が進行することを見出した。
