Outline of Annual Research Achievements

卵子は精子との受精後に全ての細胞へと分化する能力である全能性を獲得する。一方、分化した体細胞核を卵細胞質内に核移植すると、体細胞核に初期化が誘導され、クローン個体を得ることが可能である。本研究は、卵子が有する独特の性質である、体細胞核に全能性を授ける能力の分子基盤を明らかにすることを目標とする。現在のところ、体細胞核が全能性を獲得する分子的背景は多くが謎に包まれている。初期化効率の向上や全能性の包括的理解のためにもその解明の意義は大きい。
我々は体細胞核の初期化を促進する培養条件を検討する過程で、マウスクローン個体の作出効率を大幅に向上する条件を発見した。さらに、当該条件下で発現上昇する遺伝子の内、正常受精胚と比較した際に通常の体細胞核移植胚(発見した条件で処理していない核移植胚)で発現が有意に低下する遺伝子16個を同定した。この16個の遺伝子の中から体細胞核の全能性獲得に必要な遺伝子のスクリーニングを行った。具体的には、正常受精胚における個々の遺伝子の発現をsiRNAインジェクションにより抑制し、胚発生への影響を調べた。その結果、16個の内、2個の遺伝子が正常受精胚の発生に必要であることを発見した。この2遺伝子の発現様式を体細胞核移植胚で調べたところ、正常受精胚と比較して異常を示すことを見出した。核移植胚における異常な発現パターンは過剰発現によりレスキュー可能であることも発見した。これら2遺伝子についてはノックアウトマウスの作成に成功し、胚発生への関与を調べている。以上の結果より、体細胞核移植胚で発現異常を示す2遺伝子の同定に成功し、これらの遺伝子が胚発生に重要な役割を果たすことを示した。
