Outline of Annual Research Achievements

本研究は、名古屋大学トランスフォーマティブ生命分子研究所(ITbM)の理念である「合成化学と生物学の融合による新奇化合物の創出」を目指し、気孔発生に影響を与える合成化合物の探索と同定を通じて、気孔の発生ロジックの理解、操作することを目的とする。

I孔辺細胞特異的に強いGFP蛍光を発するライン(野生型背景)を用いて気孔の発生とパターン形成に影響をおよぼす化合物スクリーンを行った。スクリーニングは、誘導体の合成転換を考慮したITbM化合物ライブラリーを用いた。その結果、気孔の数を大きく増やす約100種類の化合物を得た。中でも、最も効果の高い2種類の化合物(E2とKC9)に注目した。興味深いことに、E2は消炎鎮痛薬セレコキシブのアナログである。E2は気孔の数を増やすが、その一方、植物成長阻害作用を示した。C-H活性型触媒反応を駆使して9種類のE2アナログ(構造類縁体)を合成し、その植物に対する生理作用の解析を行った。その結果、 植物成長阻害という副作用のない、気孔を増やす効果を持つ化合物の創出に成功した(Ziadi ら Chem Comm 2017論文発表)。

KC9は、気孔の数を増やし植物体の成長を促進する作用を持つ。この化合物に応答した全遺伝子発現変動を解析した。その結果、 本化合物は特定の代謝経路の発現を誘導する一方、多くの環境ストレスや病原ストレス応答は抑制されていることが判明し、KC9は植物の【成長と防御のバランス growth-defense tradeoff】に関わる経路に作用することが示唆された 。このトレードオフは、作物の増収を目指すと、その一方で耐病性やストレス耐性が減ってしまう、という育種の上でのネックとなっている。気孔の形成、代謝経路、そして成長と防御のバランスがどのような関係にあるのか、化合物の標的因子の同定を介した植物の未知の制御機構の解明が今後の課題である。この化合物は、 米国に特許出願中である。
