Outline of Annual Research Achievements

これまでの研究から、線虫温度走性における意思決定を制御するOLA-1 および DCLK-1 は多数の神経細胞に発現していた。線虫の温度走性行動は、AFD、AIY、AIZ およびRIAからなるシンプルな神経回路が主体となって駆動される(Mori and Ohshima, Nature 1995)。そこで、OLA-1が機能する神経細胞の特定を試みたところ、OLA-1遺伝子の機能細胞は複数存在し、線虫の過去の経験の違いに応じて、OLA-1は異なる細胞で意思決定を制御していることを見いだした。高温飼育された個体の意思決定の際には、OLA-1はAFD温度受容ニューロンで機能するのに対し、低温飼育された個体においては、AVK, RMG, BAGなどのこれまでに温度走性行動に関与することが知られていなかった神経細胞で働くことが示唆されていた。そこで、本年度においては、OLA-1が高温飼育された個体において、AFD温度受容ニューロンの温度応答性を制御するのかを検証した。AFDニューロンにカルシウムプローブを発現させた野生株、およびola-1変異体を作成し、カルシウムイメージングを行なったところ、ola-1変異体のAFDニューロンは、温度刺激に対して顕著な異常を示さなかった。このことから、OLA-1はAFDニューロンで機能するが、その作用はカルシウム流入よりも下流のプロセスにあることが示唆された。
また、OLA-1が、飢餓体験による温度走性行動の意思決定に関与するかを検討した。野生株の線虫は、一定の温度で餌のない条件で飼育されたのちに温度勾配上に置かれると、飢餓を体験した飼育温度には誘引されなくなる。一方で、ola-1変異体は飢餓体験後も飼育温度に誘引されることを見出した。このことから、OLA-1は飢餓体験による意思決定にも重要な役割を果たしていることが明らかとなった。
