Outline of Annual Research Achievements

次の4つの研究課題に取り組んだ。
1.ヘテロクロマチン蛋白質HP1によるメチル化ヒストン認識の動構造解析:HP1はヒストンH3K9me3を認識しヘテロクロマチン化に寄与する。本研究では、オープンな高次構造をとるジ・ヌクレオソーム環境下の2量体HP1α及びγの結合モードを解析し、HP1α二量体が、二つのヌクレオソームのH3テールとそれぞれ結合するモードが支配的であることを見出した。このモードを好む理由は、HP1αが有するリンカー領域の3つの塩基モチーフがリンカーDNAと親和性が高い点にあることが分かった。(Watanabeら, Biophys. J. 印刷中)。
2.熱揺らぎによるヌクレオソームスライディングの動的分子機構解析:単純塩基配列DNAについては、DNA長軸周りの回転と共役したスライディングが起こった。ヒストン8量体と親和性の高い塩基配列の場合は、回転共役型のスライディングとともに、回転を伴わない10塩基遷移も見られた(Niinaら PLoS Comp. Biol. 2017)。スクリュー型スライディングは、ヒストン周りのDNA1巻き(約10塩基対分)の過渡的な伸長あるいは圧縮を間欠的に形成し、それを伝搬させて進むことが分かった(Brandaniら NAR 2018)。
3.ATP依存リモデラーによるヌクレオソームスライディングの動的分子機構解析:多くのリモデラーは、ヌクレオソームのSHL2領域に結合し、ATP依存的にヌクレオソームをスライドさせている。この過程の分子シミュレーションに成功した。
4.パイオニア転写因子のヌクレオソームへの結合の動態解析:パイオニア転写因子Oct4は、ヌクレオソーム上の認識配列に結合する。シミュレーションで得られた結合様式は非常に多様であり、主にはOct4の2つのPOUドメインの一方でヌクレオソーム外側に向いたDNA配列を認識した。
