Outline of Annual Research Achievements

前年度までに、ヌクレオチド除去修復(NER)の損傷認識因子であるXPCタンパク質が、マウス細胞において自身のDNA結合活性及びヒストンH3K9me3修飾に依存せずに、pericentric heterochromatin(PHC)に局在することを明らかにした。この細胞に局所紫外線照射を行うと損傷領域とPHCが重なる部位でXPCが特に強く集積すること、またNERにおいてXPCの下流で働く基本転写因子TFIIHやXPAが通常はPHCには局在しないのに対し、局所紫外線照射領域内のPHCでは局在が見られたことから、PHCがNERにおいて何らかの積極的な役割を担っている可能性が示唆された。損傷領域がヘテロクロマチン様構造を形成する、もしくはDNA損傷部位がヘテロクロマチン領域に移動して修復されるといったまったく新しいモデルを検証するため、蛍光タンパク質を融合したヒストンを安定発現するマウス細胞株を作製し、紫外線照射に伴うクロマチン構造の動態解析を進めている。
一方、XPCがヒストンと直接相互作用し、特にヒストンH3のアセチル化がXPCとの相互作用を負に制御すること、また局所紫外線照射を行った細胞の免疫蛍光染色の結果から、H3K27ac等のアセチル化ヒストンが損傷領域から積極的に排除されている可能性が示唆されていた。マウス細胞におけるXPCの局在と合わせ、細胞内におけるXPCの局在が特に非アセチル化ヒストンH3との相互作用によって制御されている可能性を考えて、クロマチン免疫沈降シーケンス(ChIP-seq)法を用いた解析を領域内共同研究により行った。ヒト肺癌由来A549細胞を用いたChIP-seq解析の結果から、ゲノムワイドなXPCの分布と遺伝子密度との関連を示唆する興味深い結果を得た。遺伝子発現の有無やさまざまなヒストン修飾とXPCの局在との関係について、さらに詳細な解析を行っている。
