Outline of Annual Research Achievements

線虫C. elegansのAmphid sheath (AMsh)グリア細胞は頭部のニューロンを取り囲むように配置するが、老化した線虫個体ではAMshグリア細胞に空胞化や枝分かれといった形態異常が見られることがわかった。また、老化個体では温度走性行動に異常が見られることを我々は既に見出していたが、温度受容ニューロンAFD感覚末端の微絨毛構造や、AFDの温度に対する応答も個体の老化に伴って異常が見られることを新たに見出した。
一方で、グアニル酸環状化酵素(Guanylyl Cyclase (GCY)) GCY-8はAFDのみに発現し、AFDの温度受容に重要であるが、GCY-8を欠損した変異株では、老化に伴うAFDの形態異常や温度走性異常が緩和された。AMshグリアはGCY-8を抑制することでAFDの形態維持に寄与することが報告されているので(Singhvi et al., Cell, 2016)、我々が見出した結果は、老化に伴ってまずAMshグリアが異常となり、その結果GCY-8の抑制が解除されてAFDの形態が異常となり、AFDの温度応答性も異常となって、最終的に温度走性行動に異常をもたらすという可能性を示唆する。
線虫のグリアはニューロンの生存には不要なので、線虫はグリアによる神経系の機能制御を解析するのに有利な実験系である。本研究成果によって、ニューロンとグリアの機能的相互作用が老化に伴ってどのように変化するかを、線虫を用いて解析する実験系が確立された。
