Outline of Annual Research Achievements

ミクログリアの分泌するカテプシンS(CatS)欠損がシナプス機能ならびに社会的行動に及ぼす影響を解析した。(1)大脳皮質ニューロンのシナプス強度の日内変動に及ぼすCatS欠損の影響: 野生型ならびにCatS欠損マウスから作成した大脳皮質スライス標本において、大脳皮質ニューロンのシナプス強度の日内変動についパッチクランプ法を用いて解析した。野性型マウスの活動期(ZT14)における皮質ニューロンの興奮性後シナプス電流(EPSC)振幅は休止期(ZT2)より有意に大きかった。一方、CatS欠損マウスでは、活動期ならびに休止期におけるEPSC 振幅に有意な違いは求められなかった。(2)大脳皮質ニューロンのシナプス密度に及ぼすCatSの影響: 野生型マウスから作成した大脳皮質スライス標本において、リコンビナントCatSを適応しスパイン密度への影響を解析した。スパイン密度を細胞内染色した大脳皮質ニューロンより計測した結果、CatSを適応するとスパイン密度は有意に減少した。(3)社会的行動に対するCatS欠損の影響:CatS欠損マウスが社会的行動障害を示す可能性について3-コンパートメントチャンバーを用いて検討した。野性型あるいはCatS欠損マウスを中央チャンバーに入れ、左チャンバーにマウスを入れて慣らした後(Familiarマウス)、さらに右チャンバーにStrangerマウスを入れExporation timeを計測した。野性型マウスではStrangerマウスにより関心を示したが、CatS欠損マウスではFamiliarマウスとStrangerマウスに対して同程度に関心を示した。
以上の結果より、ミクログリアの分泌するカテプシンSは細胞外マットリックス分子を修飾することでシナプス強度ならびにスパイン密度の日内変化を制御し、社会的行動に関与する可能性が示唆された。
