Outline of Annual Research Achievements

透明化技術は三次元構造の高速な解析を実現するものであり、神経ネットワーク構造解明のブレイクスルーにつながると期待される。しかし、透明化された脳組織をいかに活用して神経回路構造を解析するかという『真の応用』はまだ始まったばかりである。本研究課題では透明化技術を軸に、『マクロ―メゾ―ミクロ』の各階層を有機的に結びつけ、神経ネットワーク構造を効率的に解析する方法論を確立することを目的としている。
本領域で展開されている、色素、光学顕微鏡、イメージングソフトウェア、サンプル調製を包括する技術開発を参考にしながら、平成29年度は以下の課題に取り組んだ。
(1) 大脳新皮質運動野から脊髄にまで軸索を展開する皮質脊髄路(CST)ニューロンを効率的に標識するため、順行性もしくは逆行性標識を試みた。順行性標識については、申請者らが開発した、神経細胞特異的で高発現を可能にする「AAV SynTetOff(Sohn et al., 2017)」をもとに、種々の膜移行シグナルを検討し、候補を絞ることができた。また、逆行性標識については、脂溶性カルボシアニン色素のアプライ法などを検討し、成体標本でも再現性良く標識する手法を定めた。
(2) 本領域で開発されている新規蛍光・発光プローブを、上記「AAV SynTetOff」に搭載して検討を行った。例えば平成29年度は、宮脇敦史(領域代表)らが開発した発光プローブを搭載し、その有用性を示すことができた(Iwano et al., 2018)。
(3) 透明化ステップの改変を行い、時間の短縮だけで無く、膨張・収縮を最小限に抑えるプロトコールを開発した。
(4) 光学顕微鏡観察から電子顕微鏡観察へとズームインする際に必要な、部位マーキング手法の開発を進めた。化学的・物理的マーキング法で有望な候補が複数あり、今後はさらなる検討を続けていく必要がある。
