Outline of Annual Research Achievements

1.本研究では、カタユウレイボヤ尾芽胚の尾部湾曲機構に注目し関連分子探索とその役割を解明することを目的とした。これまでに以下を明らかにした。1)カタユウレイボヤ尾芽胚期における形態的特徴量を9個抽出しホヤ尾芽胚期に相当するStage18からSt. 24までの経時変化を算出した。これらの特徴量に対し主成分分析を行った結果、第一主成分は尾部の伸長、第二主成分は尾部の曲がりを示す特徴量に分類され87%の累積寄与率を得た。2)第二主成分特異的に影響する阻害剤を探索したところ、リン酸化ミオシンを阻害するY27632およびML-7(ミオシン軽鎖キナーゼ阻害)は第二主成分のみ野生型と比較し有意差があることが分かった。3)尾芽胚期におけるリン酸化ミオシン抗体染色の結果、St. 21~23にかけてリン酸化ミオシンのシグナルが検出された。特にSt. 22の前方腹側表皮細胞の前後細胞境界に沿って強いシグナルが観察され、Y27632添加胚ではそのシグナルは消失した。局所曲率と腹側表皮の前後軸に沿ったリン酸化度合いとの相関を求めた結果、強い相関があることがわかった。4)リン酸化ミオシンの腹側表皮局在時の細胞形態を可視化するため、St. 21~24にかけて尾部の表皮細胞を3次元再構築し尾部湾曲に伴う細胞の形態変化情報を得た。一方、Y27632添加胚ではこの細胞形態変化は確認されず、細胞形態は前後軸方向へ大きく伸長していた。細胞の幾何学的な形態量を定量化し、局所曲率との相関を算出したところ、高い相関が得られた。以上より、ホヤ尾芽胚における尾部湾曲は腹側表皮におけるリン酸化ミオシンが関連する特定の細胞形態変化により引き起こされることが明らかにされた。
