Outline of Annual Research Achievements

1)SUMO E3リガーゼSIZ1によるメチル化ヒストンH3K4の調節機構の解明
SUMO E3リガーゼであるSIZ1内のPHDドメイン(C4HC3ジンクフィンガードメイン)によりヒストンH3K4me3が特異的に認識されることが明らかにしてきた。SIZ1pro:SIZ1(ΔPHD)、SIZ1pro:SIZ1(C162S)はsiz1変異体を相補できないが、SIZ1pro:SIZ1(C117S)では相補可能であった。これはC162がαヘリックスを構成しており、亜鉛との結合のみならず構造維持に関わっている可能性が示唆された。また、PHD(C162S)ではH3K4me3との結合が見られなくなるため、SIZ1pro:SIZ1(C162S)はsiz1変異体を相補できなかったものと考えられる。一方で、PHD(C117S)はH3K4me3との結合は見られることからSIZ1pro:SIZ1(C117S)ではsiz1変異体を相補できたと考えらえる。興味深いことにPHD(C117S)はH3への結合も見られ(PHDではH3へは結合しない)、この結合がどのように関わっているかはまだ明らかに出来ていない。

2)SUMO化とクロマチン制御因子との関係性およびその機能的役割の解明
SIZ1はPHDドメインを介してH3K4me3と結合したり、ヒストンH3K4メチルトランスフェラーゼATX1と結合したりと、エピジェネティクス制御に深く関わっていることが示唆される。そこで、SIZ1と相互作用する因子を単離することで、エピジェネティクス制御にどのように関わるかを明らかにすることにした。SIZ1ではtranscriptional activator、bromodomainタンパク質、transcriptional coactivator ADA2 2B、Alfin-like5 PHD domain proteinが相互作用因子として単離された。
