Outline of Annual Research Achievements

植物の主要な光受容体フィトクロムは、専ら標的遺伝子の転写量を制御することでシグナルを伝達すると考えられていたが、我々は最近、フィトクロムが転写量制御に加え、ゲノムワイドに2,000を超える遺伝子の転写開始点を変化させることにより、mRNAの5’末端の長さを制御し、その結果、数百という数のタンパク質の細胞内局在が光依存的に変化することを発見した。ここで興味深いことに、フィトクロムにより転写開始点の制御を受ける遺伝子のほとんどは、転写量(mRNAの総量)を一定に保ったまま、異なる複数の転写開始点の使用頻度をフィトクロムシグナルに応じて変化させるが、その分子機構は不明である。
本年度は、フィトクロムの転写開始点制御による光依存的なタンパク質の細胞内局在変化に異常を示す変異体をさらに多数単離し、それらについてさらに解析を進めた。また、2,000以上の標的遺伝子のプロモーター領域に濃縮される塩基配列を情報科学的に解析することにより、当該制御に関わるシス配列を同定した。そしてこれらの解析結果を組み合わせることで、フィトクロムによる転写開始点制御の分子機構についてモデルを構築した。
