Outline of Annual Research Achievements |
本課題では、環境ストレス誘導性のアンチセンスncRNAに着目し、生成に関わる新規な因子を同定し、アンチセンスncRNAを介した植物の環境ストレス適応システムの解明を目指す。
これまでに得られた研究実績は以下の通りである。 rdr1/2/6の3重変異体および野生株を用いて乾燥ストレス過程におけるトランスクリプトームを解析し、乾燥ストレス条件下でrdr1/2/6におけるアンチセンスncRNAの生成量が野生株と比べて減少する遺伝子を1,136個同定した。これら遺伝子領域の多くにおいて乾燥ストレス条件下でRDR1/2/6依存的なsiRNAの蓄積は観察されなかった。DCP5遺伝子の変異体においてアンチセンスncRNAの発現量が減少し、XRN4遺伝子の変異体においてアンチセンスncRNA発現量が増加していた。これらの結果は、センスRNAがDCP5によりdecappingされた後、センスRNAの分解産物を鋳型として、RDR1/2/6によりアンチセンスncRNAが生成することを示唆する。cordycepinを用いた解析から、rdr1/2/6変異体において乾燥ストレス誘導性遺伝子のRD29AのmRNA分解が野生株と比べて減少していることが示された。RNaseプロテクションアッセイから、アンチセンスncRNAがセンスRNAと細胞の中で二本鎖RNAを生成することも示された。rdr1/2/6の三重変異体は、野生株と比べて再吸水後の根の成長再開が遅れ主根が短い表現型を示したが、この表現型はRDRファミリーの単一変異体や二重変異体、dcl2/3/4の変異体では観察されなかった。上記の結果から、RDR1/2/6依存性のアンチセンスncRNAが既知のDCL2/3/4依存的なsiRNA生成を介さずに植物の乾燥ストレス適応に機能している事が示された。
