Outline of Annual Research Achievements

本研究では、生命科学研究を刷新するデータ駆動科学実践のモデルケースとして、データの大規模(高解像度・ハイスループット・多様)化がすすむエピゲノムデータを用いて、データから直接的にヌクレオソームレベルのスケールにおける生命現象を説明するモデルを抽出する方法論の確立を目指している。

本年度は、遺伝子発現に関わるヌクレオソームの性質を決定付ける要素について、多くの研究成果が得られた。我々が、2015年に発見した新規のヒストン亜種であるH3mm7は、不安定なヌクレオソームを形成することで、マウス骨格筋再生時の遺伝子発現を促進すること見出し、筆頭著者として論文発表を行った。共同研究においては、HeLa細胞を用いて、オーバーラッピングダイヌクレオソームとよばれる特殊な構造体(ヒストン14量体)が、遺伝子転写開始点直下に形成されることを明らかとした。また、試験管内にて再構成したアルブミン遺伝子エンハンサー領域のDNA上に形成されるヌクレオソーム配置の解析から、転写因子結合部位に対して特定のヌクレオソーム配置を形成することが明らかとなった。以上から、ヌクレオソームスケールの生命現象を解き明かすために必要な要素が徐々に明らかになりつつある。さらに、これらヌクレオソーム配置やヒストンの組成の変化と遺伝子発現の関係を明らかするため、少数細胞でエピゲノム情報を取得する技術(ChILT)を独自に開発し、論文投稿を行っている。
