Outline of Annual Research Achievements

我々はHIF-1を活性化する新規遺伝子を同定し、そのHIF-1活性化能がp53による抑制を受ける可能性を見出していた。この様な状況下、本研究で我々は、この新規遺伝子・p53・HIF-1の3者を遺伝子工学的に操作した細胞を準備し、新規遺伝子がHIF-1を活性化することによって、HIF-1下流のマトリックスメタロプロテアーゼ群の発現が誘導されることを、定量的RT-PCR法で確認した。DNAマイクロアレイによっても、この結果の正当性が裏付けられた。以上の結果を受け、当該新規遺伝子の過剰発現によって実際にがん細胞の浸潤能が亢進するのかを、ウェットの実験系で検証した。trans-well plateを用いてinvasion assayを実施したところ、新規遺伝子の過剰発現によってがん細胞の浸潤能が亢進すること、そしてそれがHIF-1αのノックダウンでキャンセルされることを見出した。また、このp70-HIF-1依存的な浸潤能の亢進が、p53ノックアウト細胞ではみられるが、野生型p53の存在下では抑制されることを見出した。
当該新規遺伝子の系統的欠失変異体を作成し、HIF-1を活性化するために必要なドメインを同定した。公開されているマイクロアレイデータベースを対象にした解析を通して、新規遺伝子の腫瘍内発現量が高い場合にがん患者の予後が不良であることを見出した。そこで、京都大学医学部附属病院で収集したヒト肺がん256症例を対象にTissue Micro Arrayを作成し、当該遺伝子を対象にした免疫組織化学染色を実施、患者の予後不良との相関を検証した。その結果、新規遺伝子のコードする蛋白質が腫瘍組織内で過剰に発現しているほど、肺がん患者の生存期間が短いことが明らかになった。
