Outline of Annual Research Achievements

不安定な概日振動能を持つ多数のニューロンの集団発振機構である視交叉上核(SCN)の中枢概日時計が、極めて安定した概日リズムを発振するメカニズムを解明することを目指している。
本年度は、AVPニューロン特異的にVgatを欠損したマウスについて、概日行動リズムの解析を終了した。AAVベクターによってVgatの発現を当該マウスのSCN・AVPニューロンにレスキューすることで、概日行動リズムが正常になることも確認した。当該マウスのSCNスライスにおいて、PER2-LUCの発現リズムの発光イメージングを行い、概日行動リズムの異常に反し、時計遺伝子発現リズムには大きな異常が見られないことが分かった。SCNスライスの電気生理学的解析で、AVPニューロンにおけるIPSCの振幅に日内変動があること、この変動がAVPニューロン特異的Vgat欠損マウスで消失することが明らかになった。さらに2,3の実験を追加して、AVPニューロン特異的Vgat欠損マウスについて論文発表する予定である。GABAに対する応答性を変化させるために、細胞内Cl-濃度を操作したマウスも作成した。AVPニューロン特異的NKCC1欠損マウス、VIPニューロン特異的NKCC1欠損マウスは共に、概日行動リズムに大きな異常は示さなかった。また、KCC2のドミナントポジティブ、ドミナントネガティブ型をAAVベクターによりSCNのAVPニューロン、VIPニューロンに特異的に発現させる実験も遂行中である。
