Outline of Annual Research Achievements

1.「サルの課題トレーニング、記録部位の同定、およびコントロールデータの取得」、2.「パーキンソン病サルモデルの作製およびモデルにおける多領域多点同時記録」、3.「多領域多点同時記録データの解析:各領域における活動変化」の3つの研究計画を実施し、以下のような研究成果を得た。
MPTP投与によって作製したパーキンソン病サルモデルから、安静時およびボタン押し課題遂行中における大脳皮質、大脳基底核、小脳から神経活動(主にLFP)の多領域多点同時記録を実施した結果、パーキンソン病モデルの小脳からベータ波の過活動を検出し、更にcross-frequency coupling解析により、運動遂行時における大脳皮質(特に一次運動野)との間のphase amplitude couplingが大脳基底核よりもむしろ小脳で顕著であることが明らかになった。具体的には、(1)時系列に基づいて、大脳基底核の淡蒼球と一次運動野との間のcross-frequency couplingを解析したところ、健常時やチックモデルではベータ帯域におけるphase amplitude coupling が運動遂行時に強く検出されるのに対して、パーキンソン病モデルでは同様のcoupling現象がほとんど消失していた。(2)同様に、小脳(主に小脳皮質)と一次運動野との間のcross-frequency couplingを解析したところ、(1)の結果と異なり、健常時やチックモデルにおいて運動遂行時にみとめられるベータ帯域でのphase amplitude coupling が、パーキンソン病モデルにおいても検出された。
以上の研究成果は、パーキンソン病の病態発現への小脳の関与を示唆しており、従来のパーキンソン病研究の範疇を超えた極めて独創的なものであるとともに、近年注目が集まっている大脳皮質、大脳基底核、および小脳の機能連関についても新たな知見を得ることができたという点で、当該新学術領域研究の推進に大きく寄与するものであると考える。
