Outline of Annual Research Achievements

(1)ゼブラフィッシュを用いて、トランスポゾンを用いた遺伝子トラップ法・エンハンサートラップ法を実施し、細胞・組織・器官特異的に改変型酵母転写因子Gal4FFを発現するトランスジェニックフィッシュを作製した。それを利用して、計画研究瀬原班と共同で、宇宙滞在によって浮き袋で発現量が亢進する遺伝子の発現をGFPにより可視化することに成功した。
(2)宇宙滞在により影響を受ける可能性がある脳領域の解析を行った。(i) 哺乳動物の扁桃体は情動や情動が関わる記憶・学習に重要な働きをしている。我々はゼブラフィッシュ終脳の特定の神経細胞でGal4FFを発現するトランスジェニックフィッシュを作製し、そのGal4FF発現神経細胞の機能を神経毒素遺伝子の発現により阻害した。その結果、そのGal4FF発現神経細胞が恐怖条件付け学習に必須であること(扁桃体に相当する機能を担っていること)を明らかにした(Lal et al. BMC Biol 2018)。(ii) 脳血管近傍に存在する新規細胞群を同定した。それらが血液脳関門形成に関わっている可能性を示した(Galanternik et al. eLife 2017)。(iii)視床下部はホメオスタシス、睡眠、食欲のコントロールに重要な働きをしている。トランスジェニックフィッシュとカルシウムイメージングを利用して、エサなどの視覚刺激が食欲中枢である視床下部下葉を活性化する神経回路を明らかにした(Muto et al. Nat Comm 2017)。
(3)計画研究瀬原班と共同で、宇宙滞在2日目、5週目のゼブラフィッシュの脳の解剖を行い、RNA-seq解析を行った。遺伝子プロファイリング解析の結果、TRPN1遺伝子を含め宇宙滞在によって発現が増減する遺伝子を複数同定することに成功した。現在それらの機能について解析を進めている。
