Outline of Annual Research Achievements

本研究では、抗腫瘍性マクロライド抗生物質FD-891のポリケチド合成酵素(PKS)の触媒ドメイン間の相互作用機構を解明し、その知見を生かしたPKS遺伝子改変によりスターター部位の異なる類縁体を生合成することを目指した。
本研究期間では、スターター部位の生合成に関わるローディングPKSのアシルキャリアープロテイン(ACP)ドメインとアシル基転移酵素(AT)ドメイン、ACPドメインとβ-ケトアシルACP合成酵素(KS)ドメインの相互作用機構を解明するために、分子プローブを用いてクロスリンク反応させた複合体を精製して結晶化を検討した。また、ACPドメインを単独発現させて、単独発現させたATドメインとKSドメインとのクロスリンク反応による複合体化と結晶化を検討した。いずれの条件においても結晶構造解析に供することのできる結晶は得られていないが、クロスリンク反応の効率が複合体サンプルの純度に大きな影響を与えることがわかった。
遺伝子改変による類縁体創製に関しては、FD-891のローディングPKSのATドメインを、前年度に生合成遺伝子クラスターを決定したビルストマイシン生合成に関わるPKSの基質特異性の異なるATドメインと入れ換えた。相同性のある酵素構造情報を基に境目を決定して入れ換えた結果、一炭素増炭した類縁体と二炭素増炭した類縁体を生合成させることに成功した。一方、メトキシ基を導入した類縁体の生合成も検討したが、期待した化合物を得ることができなかった。入れ換えたATドメインがACPドメインと相互作用できない可能性、KSドメインがメトキシ基を導入した中間体を認識できなかった可能性などが考えられる。今後はうまくできなかった原因を明らかにして、それを解消すべき遺伝子改変を行い新たな類縁体を生合成する予定である。
