Outline of Annual Research Achievements

1. Guaianolide Sesquiterpene Lactones(GSL)生合成経路の再構築:Guaianolide Sesquiterpene Lactones(GSL)は、5員環-7員環-ラクトン環が縮合した基本構造を持つ植物セスキテルペノイドの一グループであり、がん幹細胞に対する選択的な細胞毒性を発揮する化合物を含むことが報告されている。本研究では、組換え酵母内におけるGSL生合成経路の再構築を目指して、植物トランスクリプトームデータから候補となる生合成酵素遺伝子を探索し機能を解析した。シャムジンコウのセスキテルペン環化酵素、レタスのシトクロムP450モノオキシゲナーゼ、クソニンジン由来アルコールデヒドロゲナーゼおよびアルデヒドデヒドロゲナーゼを組換え酵母内で組み合わせることでGSLの前駆体となりうるδ-guaianoic acidのde novo合成に成功した。次に、ラクトン環形成に関わる酵素遺伝子の候補をシナジンコウおよびThapsia garganicaからマイニングし、計9種の候補P450の酵素機能を解析した。その結果、1種はδ-guaieneに対する酸化活性を示し13-hydroxy-δ-guaieneを、もう一種は11,12-epoxy-7-hydroxy-δ-guaieneを生成物として与えることが判明した。残りのうち4種は上記とは異なる構造未同定のδ-guaiene水酸化体を生成物として与えることが判明した。
2. ヨモギ属植物が生産する希少セスキテルペノイド生合成経路の再構築:多様なセスキテルペノイドを生産するヨモギ属に着目し生合成関連酵素を探索した。ミブヨモギから4-Amorphen-11-ol合成酵素を、A. abrotanumから7-epi-silphiperfol-5-ene合成酵素を、新規セスキテルペン合成酵素として同定した。
