地球上のほとんどの生命システムにとって分子酸素に対する細胞応答は重要な環境適応の一つであり、酸素濃度変化を検知し発現する遺伝子セットを量的・質的に変化させる。脊椎動物細胞における低酸素応答はPHD-HIF経路が最もよく知られているが、これ以外の経路の存在も示唆されている。さらに、細胞の状態に応じて酸素応答が異なることも知られており、低酸素応答システムの全貌は未だ不明である。本課題は、われわれが最近開発したiMPAQT法を中心とする定量トランスオミクスプラットフォームを用いて、いくつかのモデル細胞(正常、がん、老化など)における低酸素応答を、タンパク質発現量、水酸化、さらには代謝物の変動として包括的に計測し、それらの多階層データを統合的に解釈することで、既存の分子機構に依らない新たな生体酸素応答の実体解明を試みる。 まず、正常線維芽細胞を用いて、通常の増殖期、がん、老化、および休止期の4つのモデル状態を確立し、これを対象に絶対定量プロテオーム解析法であるiMPAQT法を用いて多数の代謝酵素の発現量の低酸素応答を時系列データで取得することに成功した。タンパク質の水酸化部位同定に関しては生理的なプロリン水酸化と実験過程で生じるメチオニンやトリプトファンなどの酸化を区別することが困難であることが判明し、これを解消するための技術開発を行った。さらに、プロリン水酸化酵素に対する結合タンパク質の同定を免疫沈降-MSおよび近位ビオチン化標識法をもちいて実施し、既存の基質であるHIF-1 alphaと共に多数の結合タンパク質の同定に成功した。
