Outline of Annual Research Achievements

ショウジョウバエの匂い連合学習では、記憶の獲得・保持・読み出しの過程を明確に区別でき、その全ての過程がキノコ体と呼ばれる単一の脳構造で処理される。本研究では、記憶の素過程を定義するキノコ体出力神経の機能パターンと、その遷移を理解することを目的とする。研究代表者らの研究グループは、キノコ体の出力を担う21種類の出力神経を全て同定した。つまりキノコ体は、全ての神経種が遺伝学的に標識でき、機能操作と行動出力との因果関係を明らかにすることができる独自のモデル回路である。本研究ではこの遺伝学的リソースを用い、キノコ体の出力を構造と機能の両面から解析する。
2018年度は、キノコ体の異なる記憶の素過程における出力パターンを調べるために、嗅覚報酬・嗅覚罰記憶の各素過程における各出力神経の必要性を定量した。全ての出力神経について、その神経伝達を各素過程特異的に阻害し、記憶学習実験を行った。各出力神経の記憶に対する阻害効果から、各素過程での貢献度を定量することで、キノコ体からの出力コードが時間とともに変化する様子を明らかにした。さらに、各素過程の機能コードを実際の顕微鏡画像上にプロットし、その出力先、放出される神経伝達物質を可視化した。各出力神経細胞種の形態的特徴と記憶の各素過程の機能コードを統合することにより、例えば報酬記憶の獲得に共通する神経回路モチーフを同定した。
