Outline of Annual Research Achievements

快情動行動が発現する際、ドーパミンがD1R-PKA-Rap1-MAPK経路を活性化させることで、側坐核のドーパミンD1受容体発現中型有棘神経細胞(D1R-MSN)が興奮性の低い状態から高い状態へとシフトし神経回路が作動し快情動行動が発現する(Nagai et al. 2016)。
これらの細胞内シグナル伝達機構の役割を分子レベルで観察・操作するために平成30年度は以下の研究を実施した。
1) グルタミン酸(NMDA)、アデノシン受容体作動薬でリン酸化されるリン酸化基質および変動するリン酸化部位を同定した。2) Rho-キナーゼのMYPT1のドッキング部位とドッキングモチーフを同定した。3) 側坐核におけるD1R-MSNおよびD2R-MSN特異的なドーパミンおよびアデノシンの作用機序を解明した。D1R-MSNからD2R-MSNへ、またはその逆への活性シフトが、主にドーパミン濃度の変化に依存するというモデルを提案した。D2受容体拮抗薬がアデノシンシグナルを増強して作用することを見出した(Zhang et al. Neurochem Int 2019)。4) MAPKによるKCNQ2のリン酸化部位に対して抗リン酸化抗体を作製した。D1R作動薬で刺激した線条体スライスにおいてKCNQ2のリン酸化が亢進し、このリン酸化亢進はMAPK阻害剤の処置で抑制された。5)KClやNMDAで刺激した線条体スライスにおいてShank3のリン酸化が亢進し、そのリン酸化の亢進はRho-キナーゼ阻害剤の処置により抑制された。また、Shank3のリン酸化により、Dlgap3, PSD95, 14-3-3, NMDARとの相互作用が増強されることを見出した。6)ドーパミンがD1R/PKA/MAPKシグナルを介して、Npas4などの転写因子をリン酸化し、報酬学習・記憶を制御することを明らかにした。
