Outline of Annual Research Achievements

大腸がん発生過程では、遺伝子変異の蓄積により段階的に悪性化進展する「多段階発がん」の概念が確立している。一方で、「細胞競合」の概念では、遺伝子変異した細胞が周囲の細胞に排除されるため、多段階発がんとは相反する生物現象である。本研究では、大腸がんドライバー遺伝子であるApc、Kras、Tgfbr2、Trp53(A、K、T、Pと略す)に異なる組み合わせで変異を導入したマウス腸管腫瘍細胞を用いた細胞競合実験を計画し、下記の実験を実施した。
(1)異なる遺伝子型腫瘍の混合移植実験:AKTP細胞をマウス脾臓に移植すると肝転移するが、A、AK、AT、AP細胞は転移能を獲得していない。そこで、AKTP細胞をGFP(緑色蛍光)標識、それ以外の腫瘍細胞をTdTomato(赤色蛍光)標識し、1対1の割合でAKTPと他の細胞を混合して脾臓移植実験を実施した。その結果、ATまたはAP細胞とAKTP細胞を混合移植した場合、80%以上の肝転移巣はAKTP細胞で構成されたが、それ以外の転移巣ではAT、AP細胞が混合して生存し増殖した。一方で、AおよびAK細胞はAKTP細胞と混合移植しても転移しなかった。
(2)混合転移巣におけるAKTP細胞枯渇実験:次に、AKTPとの共存により転移したATおよびAP細胞による肝転移巣形成が、AKTP細胞に依存しているか明らかにするため、AKTP細胞にジフテリア毒素受容体(DTR)遺伝子を導入し、転移巣形成後にマウスにDTを投与してAKTP細胞を枯渇させた。その結果、AP細胞はAKTP細胞非存在下でも増殖して転移巣を形成したのに対し、AT細胞は生存しなかった。
以上の研究成果から、がん細胞の悪性化過程では細胞競合による排除機構よりも、多様性の維持による悪性化誘導機構が働いていると考えられた。一方で、多様性による悪性化誘導は腫瘍細胞の遺伝的条件に依存していると考えられた。
