Outline of Annual Research Achievements

私たちはこれまでの研究から、細胞競合を引き起こすようなRas, Myc, Srcなどのがん遺伝子に変異がおこった老化細胞は、正常な細胞にくらべてはるかに多くの蛋白質・核酸・分泌膜小胞・脂質などのSASP因子を細胞外へ放出していることを明らかにしている。そこで北海道大学遺伝子病制御研究所の藤田先生との共同研究によりMDCK(イヌ腎臓尿細管上皮細胞)を用いた細胞競合の誘導システムを導入し、老化細胞が分泌するSASP因子のうち細胞競合を制御する分子のスクリーニングを行った。その結果、老化細胞が分泌するSASP因子によって細胞競合が抑制されることを見出した。さらに、活性化型Rasによる細胞老化とDNA損傷による細胞老化を誘導する前後のヒト正常二倍体細胞の培養上清のサイトカインアレイ解析を行い、SASP因子のうちで老化した細胞に共通して分泌が亢進している因子を約10種類同定した。それらの候補因子の中から、それぞれ1種類のSASP因子を添加した状態でMDCK細胞を用いた細胞競合解析を行ったところ、あるSASP因子の添加によって細胞競合現象が阻害されることを見出した。また、この因子は培養上清に添加した時だけでなく、コラーゲンコート中に含まれた状態で機能することも明らかとなった。さらに、私たちが本研究で見出したSASP因子が生体内で細胞競合を抑制し発がん頻度の上昇に関与しているかどうかを明らかにするために個体で細胞競合を誘導することのできるマウスモデルを導入し解析を行っている。
